Probably not: If you have swelling, point tenderness on the joint line, and decreased range of motion, you need to see an orthopedic surgeon. Could be a meniscus tear. Running will only exacerbate your symptoms and could cause irreversible damage to your knee.

Could be serious: You should see your doctor soon. An MRI may be indicated to evaluate for ligament or tendon injury. I would caution against the marathon until this has been thoroghly evaluated.
