It might work, but not necessarily. In the US, about half of all syphilis is caused by strains resistant to azithromycin (Zpack). Clindamycin is active against Treponema pallidum, the bacteria that causes syphilis, but its effectiveness in treating syphilis has not been studied; the necessary dose isn't known, and it should not be relied on to abort syphilis.

Treatment for Syphilis includes Penicillin, Doxycycline 100mg twice a day for 14 days, Ceftriaxone 1 g injected intramuscularly or Azithromycin 2 grams orally. A Z-pack (250mg) would not be enough, even if taken over the full 5 days, and may result in resistance. Better to get tested difinitively for Syphilis and get appropriate treatment, than wait and have it be worse with time.

The active ingredient in a Z-Pak, if taken for the proper dosage and length of time, would treat syphilis. Clindamycin is not among the antibiotics that we used to treat that.
