Generally no: The blink reflex tests two cranial nerves, the fifth which mediates the sensation from the cornea to the brain and the seventh which causes the eyelids to blink. If the cause of the headaches is affecting those nerves, then yes it can be helpful. Most likely it will not provide the diagnosis though.
