Venous thrombosis : Prolonged sitting can increase risk of clots in legs. These clots can move into the lungs (pulmonary embolism), a serious condition. The altitude would be an additional burden due to low oxygen levels.He probably will need to be on blood thinners as well which will require monitoring. Please get clearance from his doctor before going on this vacation.

Yes: Your father should take the following precautions: if he decides on traveling by car, he should take periodic breaks every hour to stretch his legs and circulate the blood in his legs. If he has a known lung or heart problem in association with these blood clots, than yes, the altitude could pose a small problem with his breathing.
