Possible but likely: Harmony is testing fetal dna thus giving a much more accurate result. First trimester screening is a good test but works differently. It tests your hormone levels along with a sonographic measurement of the fetal neck. These are different technologies to assess risk.

Not likely: Your numbers are quite good. The 1 in 3000 is based on your first trimester screen which is a probability. The harmony test is based on the fetal dna in your blood. Therefore, it has a higher detection rate than the first trimester screen, and in theory could change--but not highly likely given your good numbers.
