No: Both medications are considered decongestants, so you only want to take one or the other, not both. If you take the sudafed, i would take it about 30 minutes prior to flying. If you use the afrin, (oxymetazoline) it tends to start working fairly quickly, so 5-10 minutes prior to flying should be adequate.
