Cultures: Usually if you have a sore throat, you should have the tonsils swabbed for strep infection. However you may ( and should) also get tested for mono as well as some blood tests depending on the severity of your illness.

Depends: In most cases a quick strep preformed on a throat swab will be all that is needed if positive. If negative some might do a backup culture. If the pattern of your symptoms suggests a more involved process, a blood test for mono may be done, but these can be falsely negative early in the illness.
