Depends on drug: Insulin works right away. With other drugs, the earliest approx time before you may expect to see a lowering of glucose is: sulfonylureas (eg glyburide); 2 days; metformin: 3-7 days; sitagliptin: 2 weeks; exenatide: 1-2 weeks; pioglitazone: 3-4 weeks. Note: these approx times assume you are checking your glu daily and you may notice a trend. It takes 2 months to see any difference in a1c.
