Infection ?: If you have an infection, then the penicillin will start working within the first 24 hours. If you do not have an infection, the penicillin will do nothing for you, since it is an antibiotic and not a pain med. See a dentist to be treated.

Dentist needed: Unfortunately antibiotics alone won't cure a toothache if it's due to an abscess. A dental exam is needed.

Usually 24 hours: Please keep in mind that antibiotics can only help reduce an infection caused by a decayed tooth, they can not eliminate the source of the infection. You must have the infected tooth treated by a dentist in tooth pain order to solve this problem. Also if your toothache is caused by a cavity, but the tooth is not accessed, an antibiotic will not help.
