No: No, you do not. Diabetes is usually diagnosed by your blood sugar levels, although someone with high blood sugar levels, especially if very high and newly diagnosed, will typically have glucose and ketones spilling over into their urine anyway, but that isn't typically how diabetes is diagnosed.
