Depends: There are no routine tests. It depends on the types of tests to be done as various blood constituents need different types of preservatives and hence different colored tubes. Generally about 10 ml is sufficient.

Many different cause: Fatigue and exhausted feelings can be due to many different reasons. If there is no physical abnormality you should look into emotional issues like depression or lack of sleep due to anxiety, depression or breathing problems like sleep apnea which can rob one from a restful sleep leading to a tired exhausted feeling next day.

Cytosis: Erythrocytosis and leukocytosis mean elevation of the rbc and WBC counts respectively above the reference values of the lab. Slight increase could be physiologic reactions but usually when leukocytosis is accompanied by fever, it signifies infection. In your body. Elevation of rbc or WBC with many persistent abnormal young forms could mean early dysplasia or leukemia.
