Depends on the time: If you took two 500 mg tablets, then you need to wait at least 4-6 hours before taking anymore, regardless what strength the other ones are. Also, you should watch the amount of Acetaminophen (that's the 500 mg or 325 mg you're talking about) you take, as you do not want to exceed 4000 mg in a 24-hour period.
