No: As long as the medications are still keeping your numbers in check, there is no long term side effects. Glimepiride loses effectiveness over time requiring a change in regimen but if it's still working for you, then no harm done.

Potentially, but...: There are potential side effects of all medications -- but the side effects of poorly controlled diabetes (because you didn't take your medication properly) are millions of times greater and possibly fatal.

Each patient unique: Long-term metformin use can cause vitamin B12 deficiency, which leads to anemia and nerve symptoms in hands/feet. Some patients also develop GI symptoms (diarrhea, nausea) even after tolerating metformin initially. Long term sulfonylurea use (such as glimepiride) can cause weight gain, hypoglycemia and faster burnout of pancreatic insulin. See a specialist about which drug regimen is best for you.
