About 6 hours: Half life of Amoxicillin is only about an hour; in general, it takes about 4-5 half lives to clear a drug to undetectable levels, so 6 hours or so is adequate.

6-8 hours or more: The half-life of Amoxicillin is 61.3 minutes. Approximately 60% of an orally administered dose of Amoxicillin is excreted in the urine within 6 to 8 hours. Detectable serum levels are observed up to 8 hours after an orally administered dose of amoxicillin. These numbers can vary from individual to individual based upon a numbe of factors.
