NO IT CAN NOT: Prednisone and other steroids can not reverse the beneficial effects of vaccines.

Depends: If you are on high dose prednisone at the time of a vaccine or in the days leading up to it, the immune system may "ignore" or not respond to the vaccine. I prefer to wait at least 4 weeks after high dose steroids to give any vaccine. Prednisone would not cancel an already established immunity from a vaccine.
