Historically this term has referred to celiac disease which is a condition that results in abdominal pain, bloating, changes in stools, weight loss, fatigue, and weakness when someone with this problem consumes foods with gluten.

Wording issues: You will be intolerant to gluten if you are allergic to it.Your body will have reactions to it and many could be severe. Allergy skin tests and blood tests would be positive. You could be intolerant to gluten but not have typical allergic reactions (hives, difficulty breathing, positive skin and blood tests), but you might have indigestion, constant bloating, intermittent diarrhea, etc.

Wondering what kind of reaction you are referring to, the time course, symptoms, etc. There are many different reactions to foods including true allergic reactions, food intolerance, or immune-mediated inflammation to gluten in people with celiac disease. Antibodies against tissue transglutaminase (anti-TTG), which is highly sensitive and specific is used as a screening tool.

You can't definitive: The only way to make an accurate diagnosis is with blood tests or an intestinal biopsy.  Many people will feel better with gluten avoidance, even without gluten sensitivity so this is not and accurate method of diagnosis.
