Depends on concern..: The most common cause of what you describe is an infection. If rarer, more serious things need to be ruled out because the symptoms are not quite fitting, then there are more specialized (though somewhat invasive) tests to look for a leukemia or lymphoma. Performing a biopsy of an enlarged lymph node, or a bone marrow biopsy can help rule out cancer. I hope he feels better soon.
