No: Lymph nodes, even when enlarged during an infection, are not necessarily painful. The key is making sure enlarged lymph nodes return to their normal size, usually within 1-3 weeks of an infection.

Lymphadenopathy: Swollen lymph nodes or lymphadenopathy is often a benign transient condition caused by reactions to acute and chronic infections. This reactive lymphadenopathy triggers a cascade of inflammation that makes these nodes tender, enlarged and rubbery in consistency. It would be helpful to know where the nodes are. Non tender lymph nodes may be an ominous sign of lymphoma, autoimmune disease or cancer.
