DVT: It is possible to develop if you have a tendency to it. It is also possible to just have a cramp from leg position and tightness from driving. You could see a dr. And get it checked to be sure. If it goes away very quickly more likely a cramp but still won't hurt to be checked.

It could, but rare: In a 28yo without other medical issues or bleeding problem to predispose u to blood clots this would be very unusual. Additionally, that's not a very long car ride to have stasis of the blood pooled in your leg veins. U likely have muscle cramping due to dehydration, low magnesium levels, or something else. If the pain worsens or u develop swelling or warmth of your leg call your doc to eval.

Possible: Dvt is also often associated with redness and swelling and you don't describe that. Also isolated calf dvt, even if you have it, only moves up the leg 20% of the time. You are young (and assuming you have no other illness that might promote dvt) i'd give it a couple of days before seeing your doctor to see if the cramp improves.

Very possible: Any calf pain or swelling after a prolonged sitting could be indicative of a dvt. In medicine we have very little reason to ignore calf pain without getting a doppler study to rule out a blood clot.

Possible: Any prolonged immobility (we typically use 2 hours, but it can be shorter) can cause dvt. If you smoke, or have any other DVT risk factors, your risk goes up.

Dvt: Dvt is possible and if concerned a venous Doppler sonogram will clarify It is very likely that this is a muscle related issue
