Can long-term tramadol use (2 yr, 300mg) cause acalculous gallbladder disease?

Unlikely: Tramadol does give frequent gastrointestinal side effects such as dyspepsia and nausea, occasional diarrhea, and others. Causing acalculous cholecystitis is not documented. An ultrasound and hida scan will be helpful in diagnosis of the disease.
