Fast/2 hr postmeal: Fasting and 2 hr post prandial (post meal) are the best time to check blood sugars as there are normal ranges used to assess these levels.

AM and PM: If you doctor wants you to check your sugars daily, you should at least check first thing in the morning (fasting) and 2 hours after your evening meal. Theoretically this will give the highest and lowest sugars of the day. If you take meds or shots with meals, checking sugars before each meal is also important.

Doctor: It is important to check fasting blood/plasma glucose. You should eat a normal diet and be fasting for at-least 8 hours. You should have your doctor arrange a test at a reputable lab. Blood/plasma glucose levels drop on storage of blood, so blood should be tested immediately. Finger stick tests with glucometers are not comparable to plasma glucose done is a lab.
