How can my husband know if he got brain cancer from his chemo on cancer brain metastases?

Chemo didn't do it: Sadly, metastatic lesions to the brain are not at all unusual. It is a frequent event with a cancer that was not localized on detection and had already spread to other areas. That is why chemo was started. Sorry for your husband's situation.
