Depends: What is the pathogen, what type of infection are you referring to? The major defense mechanism is the integument (skin and mucous membranes). Following that you have antibodies, fixed tissue immunity, phagocytic cells, and so on and so on.

Several...: It has to get into the body, so either through the skin, the intestines, or the lungs. Once inside, it has to get past the immune system with the white blood cells.
