At implantation: If an embryo is floating towards the uterus, there isn't any HCG to go into the woman's bloodstream yet (thus no rise in HCG in her blood and urine). At implantation, the HCG can now get into the woman's bloodstream, and after a few days will be at a high enough level to be detectable. A urine pregnancy test done correctly will give a reliable result the week after missing a period.
