That depends: Some employers use "point of care" (POC) testing, and this can be read within minutes of the test.  If it is all negative then the results are available within those few minutes. However, if one or more of the tests are positive, it is generally sent to the reference lab for confrimatory testing, and this can takes several days or even a week or more, depending on how busy the lab is.

Second hand smoke: Second hand smoke from weed is more likely to cause you to have a positive urine test for THC than any transmission through semen. I doubt the explanation of people blaming semen for positive drug test.
Have a healthy diet, exercise 30 minutes/day, drink plenty of water daily so your urine is mostly colorless, have safe sex, no tobacco alcohol weed or street drugs. 
Get HPV vaccine.

: Amitriptyline is not commonly included in urine drug screens or not tested for. Sometimes amitriptyline can cause a false positive for LSD, but as long as the individual being tested informs the testing entity about all drugs being taken as prescribed, it cannot be held against the patient.

Urine drug test: This depends what kind of test. Certain uds (urine drug screens) done in an office settings are usually immediate. Confirmatory uds require more sophisticated instruments such as high performance liquid chromatography may take less than one week.

No: Nicotine isn't tested for on the five panel test, but are you concerned about nicotine? I take it, you have a DOT safety sensitive job. You might not be allowed to smoke at work, but you didn't violate for nicotine. You violated for another substance, if you have already been referred to a SAP-Substance Abuse Professional. The test shouldn't take long before you will have the results.

Benzodiazepines: Drug testing can be positive for benzodiazepines for weeks after use.  It gets worse the more you have taken or the longer time you have taken them. But, it can be six or so weeks till your urine test do not return positive for benzos.

NO: NO- not related unless drugs were used during sexual relations.

Accepted Answer

No: There are a few non-steroidal anti-inflammatory medicines that can cause false positive tests for benzodiazepines -- and sometimes zoloft (sertraline). But there are no diabetes ones that i know of.  Confirmatory tests would be run for any questionable results.
