Not usually: A small amount of fluid in the cul de sac is normal.  The abdominal/pelvic cavity is moist with peritoneal fluid and when standing or sitting gravity causes the fluid to pool in the cul de sac.  Larger amounts can be seen with a ruptured cyst and will usually resorb on it's own.

Fluid can accumulate from many causes, and can be seen intermittently due to your menstrual cycle or indicate inflammation in the pelvis.

Fluid in cul de sac: This is often seen when doing an ultrasound for abdominal pain in females.  If no signs of abscess or appendicitis then in can come from a rupture ovarian cyst.

Not related: Free fluid in the cul-de-sac does not relate to what is going on in the uterus.  See your doctor for a lab pregnancy test if things remain unclear after your ultrasound.
