Sorry for your frustration but without seeing the lesion up close, it is extremely difficult to help you, especially three doctors have failed to make the diagnosis. Because the lesion is painful so it is for sure not consistent with vestibular papillomatosis which is a common, benign, asymptomatic lesion of the vagina. Please seek a fourth opinion from an experienced GYN doc or a dermatologist.
