Depends: Onset of action of a pill will depends on the amount of time it takes for the active ingredient of that pill to enter into blood stream. It will depend on time it takes to absorb from GI tract and sometimes metabolism in liver. Some drugs absorb in active form will act quicker than the drugs that need metabolism in liver.

Usually immediately: For the combined oral contraceptive pill (this is the standard form of 'the pill'), you will be protected from the day you start taking it if you start on the 1st day of your period.
