It depends: Since I have no idea what is being tested i can not give you a good answer. If, for example, salivary cortisol levels are being tested, yes, the results won't reflect your normal state of being but will show how your body reacts to a lack of sleep. It's not clear to my why you have to take it today. It's ideal to wait until a "normal day" but if you do it today your doc should know what happened.
