Mutations: Just like humans, germs have dna that occasionally mutates. For some mutations it will lead to development of new cell wall structure or other features that alter the germ enough that a specific antibiotic no longer works to kill it. The more often an antibiotic is used in the general population, the more pressure exists to create a resistant strain.
