Possibly: The timing of the positive test depends on when you ovulated and when you possibly got pregnant. If you have a regular 28 day cycle and ovulate on day 14, then yes, a home urine pregnancy test should be positive at this time. I you have a regular 35 day cycle and ovulate on day 21, then maybe no, the test would not always be positive by now.

Check again 1 week: It should be positive now if your periods are regular, but to be safe if no period in one week would repeat pregnancy test.
