Yes: Many pain medications like Codeine are pro-drugs which get converted in the liver to an active drug like morphine and other metabolites. Some people have genetic enzyme deficiencies and can't convert these drugs into an active form and have minimal to no pain relief. Ask your doctor about other medications, preferably non-opiod.
