Cleaning an out building when i pulled cardboard box off top shelf and it was full of rain water from the leak in the roof, some of the water on top of the box got in my nose could i get naegleria fowleri from this ?

Would not be surprised if your water-filled box contained bacteria and/or molds or fungi, BUT would be amazed if amoeba could survive in that milieu. Since you appear anxious, go to an urgent care center, or see your doctor and get your concerns addressed.
