I took lactolose and it was about two teaspoons and it only took about 5 hours to kick in and work is this normal as it says it usually takes 48 hrs?

You may be confusion different aspects and reasons for lactulose treatment. As a laxative, it works very quickly, often before 5 hours. The other main use of lactulose is to help control certain symptoms of advanced liver disease (which it does largely by working as a laxative). Improvement in those symptoms often takes 2 days or longer.
