If you are vaccinated, your body has a backup supply in the form of B cells; on being exposed to an infection, these will be activated and as plasma cells, produce a burst of antibodies (to spike protein).

Testing for antibody after vaccination is not recommended and is not needed. Your body will make more antibody as it encounters viral antigen, either as viral infection or as booster dose of vaccine.
