In about 24 hrs: as half life of simvastatin is 5 hrs , means half of it is eliminated in 5 hrs , after 24 hrs most of it eliminated and ready for next dose and cumulative effect will be minimum .

24-48 hours: The listed half life is 4 hours. That means after 4 hours 50%is gone. After 6 half lives (24 hours) over 96% is gone from your system. However, side effects from simva may take more time to resolve. Eg. 2-4 weeks for muscle aches. Hope this helps

If you are having muscle aches in particular you may want to try the supplement CoQ10 which helps a lot of people with Statin related muscle aches or pains.
