Usually not: The usual urine pregnancy tests are intended for a few drops of urine. If more urine is placed in the "test well", the test paper underneath may get a bit more soaked, but the urine should stop flowing through the test paper after it is soaked with urine. Maybe in a few cases, a negative test might turn positive if a few more drops had been placed, but it's better to just retest after 1 more week.
