I've had a headache for two days straight and have taken pain killers but the headache remains, could it be from the weather?

Possibly: Have you reacted to weather in your past? If so - sure! If not, still perhaps if there have been unusual conditions or if you are changing in some way. Don't overuse the pin killers if they aren't working. Try environmental impacts - change heat level, dryness level, drink water, sinus drainage efforts - positional, steam, etc. Neck massage may help. Call doctor if it goes into week.
