Mononucleosis: Yes. Very young children who have mononucleosis often test negative for the the ilness. However, in older children especially adolescents, the test is always positive if they have mononucleosis.

Depends on test: The so-called mono spot or heterophile test for mono can be negative during the first week of illness. The specific ebv antibodies can be negative at onset but will show typical changes with a week or so unless the mono is not ebv related such as todo or cmv.

Yes: The test done may not be positive early in the course of your disease. Also, most screening tests have high sensitivity in picking things up but nothing is 100%. If clinically you have the symptoms the medical provider will let you know if that is what they think you have. Good luck.
