Pelvic exam: If you are concerned about a vaginal infection, a gynecologist can use a speculum to assess the discharge, look under a microscope and send it for culture if needed. Based on the findings, they may opt to empirically treat you as well.

Vaginitis exam: The gynecologist will do an internal pelvic exam and take some cultures from your cervix an vaginal. Also, most will do a smear to look for things like yeast and clue cells to diagnose the most likely type of vaginal infection to treat.
