Biopsy: The diagnosis of pemphigus is made by history and examination of characteristic lesions on the skin or in the mouth and throat. A definitive diagnosis is made by biopsy with examination of the tissue by a dermatopathologist.

Skin Biopsy w/ DIF: Diagnosing pemphigus vulgaris (and for that matter all autoimmune skin blistering diseases) requires two skin biopsies. One is a regular biospy (h&e) and the other is an immunofluorescence test (dif). The dif is a very special test that is very specific for these diseases. It is a must to accurately diagnose the condition. Blood tests can be used to help in diagnosis and monitor treatment.
