No: The idea of a vasectomy is to block the sperm. Usually men are tested to ensure there are no sperm in the ejaculate. If you want sperm you may need to consider a reversal procedure. There is no reliable way to check for sperm at home and based on the fact he has had a vasectomy, he shouldn't have any.
