Depends: For lab test known as ELISA, the primary antibody binds to the antigen in question and the secondary antibody binds to the first antibody. It is a technical detail with little significance if you are not working in a lab. If a physician uses these terms with regard to immune processes, s/he may be referring to IgM antibody as primary and IgG as secondary. In the immune response, IgM comes first
