Yes it can: A blood test after a week of syptoms should be positive if you have mono, but there are also non-mono, viral like illnesses that will give you symptoms of mono and a negative test.

No: Mono goes throughout the body and not just one tonsil. One tonsil can be larger than the other with throat infections, including mono. Rocephin (ceftriaxone) is very powerful and should clear up almost any bacterial tonsillitis. Follow up with your doctor if you are not improving.
