Either: Both methods are used. However, it is generally accepted that tracing the endocardial borders of the left ventricle at end systole and end diastole in two separate planes gives the best estimate of ejection fraction. When using only the m-mode tracing, there are many assumptions built into the estimate of ejection fraction that can challenge accuracy.
