Blood test: If you were pregnant, at three weeks after a missed period you should an unequivocal positive result on urine pregnancy test. You may wish to get a blood test for HCG level which would be conclusive one way or the other.
If you do not wish to be pregnant, use contraception all the time, every time. You may consider implanted contraceptive, or IUD. 
Practice safe sex. 
Get HPV vaccine.

Pregnancy test: Well, if your first one was negative and the second was positive and you had missed period, most likely you're pregnant. If in doubt, do a 3rd one.

Congratulations: You're most likely pregnant.  A false negative is much more likely than a false positive.  Either see your doctor for a confirmatory test or wait a few days and retake the pregnancy test.  
False positives can happen under certain circumstances but are very rare.

Positive: Even a faint positive is a positive test. Go see your primary care physician or OB/GYN soon for prenatal care. If this isn't a desired pregnancy the earlier you seek care the more options you have. If this is desired, congratulations! Start taking pre-natal vitamins immediately if you are not already. If you retest in a few days then the line will likely be darker as the hCG levels increase.

See below: Every brand of pregnancy test is different but usually there are two possible lines. One is the control line and should always come up. The other is the test line. Usually any line in the test area is considered a positive. If you aren't sure there are tests that give an answer in the form of "pregnant" or "not pregnant" so you might want to try one of those.

Probably nothing: Menstrual irregularities are not uncommon and anovulatory cycle is a common cause.You may wish to repeat hpt in 3-4 days to make sure that you did not test too early. Yes…: You can trust a negative blood pregnancy test. There are many reasons why a menstrual cycle does not occur including not ovulating during the cycle. However, anxiety alone can cause this to occur by affecting the normal hormonal axis.
