You should: Am not aware of people preferring blood tests to urine tests for cotinine, particularly since it can be found in blood only for a few hours after smoking, while it persists in urine for several days.  In any case, testing should be negative 4 days later.  More important than passing the test is actually quitting smoking, and taking care of your health.  Please quit and stay quit!

Likely: Six days should be enough to pass the test. If you can make it 6 days without smoking do not ever do it again.
For good health - Have a diet rich in fresh vegetables, fruits, whole grains, milk and milk products, nuts, beans, legumes, lentils and small amounts of lean meats. Avoid saturated fats. Drink enough water daily, so that your urine is mostly colorless. Exercise at least 150 minutes/week and increase the intensity of exercise gradually. Do not use tobacco, alcohol, weed or street drugs in any form. 
Practice safe sex.

Likely: Three weeks would be sufficient time to clear metabolites of nicotine. You know that you need to quit smoking permanently.

Stop for 2 weeks: To be safe, you might want to stop smoking for a couple weeks.  They are testing for cotinine which is a metabolite of nicotine. It has a long half life up to 40 hours. This tests shows positive for both active smokers and those who have quit but are using nicotine patch, gum, etc. Another test is used to identify active smokers. It is called anabasine and found in tobacco products. Try 2 wks off.

Yes: If they are specifically testing for it.  Cotinine, the main metabolite of nicotine, is generally tested for in urine, where it is present in larger amounts than in blood and is easier to find.  However, it can be detected in blood as well.  If you are a smoker, the easiest way to "beat" the test is to stop smoking!  please consider stopping and getting help doing it if you need to.

Depends when test: I s done.  If done within a few minutes or perhaps an hour of that puff, it might show up.  If done many hours or days later, it should not.  Most testing is done through urine rather than blood, because nicotine metabolites can be detected in higher amounts and for longer periods of time in urine.

Impossible to say: First, am not sure anybody is doing blood tests for nicotine or cotinine, because neither stays in blood more than a few hours. Urine testing is the gold standard, and remains positive for about 2 days after a single exposure. There is nothing healthier about using a hookah, and you should strongly consider discontinuing your tobacco habit before it harms your health, rather than fear of a test.

No way to know: What you mean.  The method by which you smoke, whether cigarette, pipe, cigar or a hookah does not change the amount of nicotine you take in.  Unless you drank the water from the hookah, whatever nicotine you took in was in the smoke, and depends on how much you smoked. Odds are your test will be positive.

If you are being: Tested, and you know you are being tested, then why would you smoke a cigarette?  Most insurance companies and others who test for nicotine use urine rather than blood, but in any case a single cigarette would give you metabolites in blood for a handful of hours, and at most a day.  You need to look at your impulsivity and judgment if you get tested regularly and still do it. Counseling might help.
