Pimple?: Due to the limited space in the ear canal and sensitivity of the skin on the outer ear, minor issues can cause a lot of pain. Should get it checked to see if can relieve pain and if additional treatment needed (your dr / urgent care).

Unlikely: You should show your bump to your primary doctor. 
Fine needle aspiration is the fast informative procedure for fast answer. Most probably you have benign lesion and you have migraine not from lesion, just because you think about this lesion constantly.
Doctor's examination needed. It could be inflammed cyst ( thyroglossal, for example).

Have it looked at: It's difficult for individuals to treat bumps in the ears.  An ear nose and throat surgeon (ent), may be the best choice to treat those areas.  If it is a "zit", an oral antiinflammatory like Ibuprofen may help, as could topical benzoyl peroxide (avoid getting on the ear drum).  Bottom line, have a physician examine this area and don't try to treat it yourself.

: What you are feeling is likely a normal structure, the mastoid process. If the "lump" double in size, becomes painful, ulcerates, bleeds or other lumps appear, see your doctor.

See your doctor: You should see a doctor right away.  There are many causes for sore throat and some require treatment with antibiotics.  This includes strep throat and more rare conditions such as an abscess beside a tonsil.  Other causes of sore throat are viruses including mononucleosis.   A doctor can determine what kind of treatment is needed.  You could also get dehydrated if you aren't able to swallow.

Doctor can examine: Bumps are hard to figure out without an exam. Sometimes, they appear and disappear for unknown reasons. Bumps may be ok if there is no pain, oozing, bleeding, itching, etc...  Otherwise, the causes may be irritant or allergic (something touched the area), or may be from blocked pores, blocked glands, irritated follicles, etc...  A doctor can look at the bumps to help get a diagnosis.

Possibly darwin bump: Some people have a small bump on the inside of their upper ear. This is known as "darwin's tubercle, " or "darwin's bump, " because charles darwin mentioned it in his book. Genetics determines it presence. It is present from birth if not this, then not sure from description. Physician could tell you by physical exam.
