Some yes, some no: Herpes often is carried for many years before it is transmitted; many cases come from monogamous partners. Some STDs can be present for many years and then show up with new symptoms or lab test, like an abnormal pap smear with HPV -- seems like a new infection but isn't. Sometimes a positive test is wrong. New gonorrhea or chlamydia always means one partner has had sex with someone else.
