Drug test: You can take a urine drug test. Vyvanse (lisdexamfetamine) is Dextroamphetamine and will show up as 'amphetamine' on a drug test.

You don't, generally: Standard "drug tests" will detect recent vyvanse (lisdexamfetamine). It will show up as an amphetamine. Blood tests are not done, except perhaps for forensic reasons.
